Cobb spielte zunächst Klavier und Violine und später Tenorsaxophon. Er wurde 1933 Berufsmusiker und begann seine Karriere als Mitglied der Bands von Frank Davis und Chester Boone. 1936 wechselte er zu Milt Larkin, bei dem er sechs Jahre blieb. 1942 wurde er der Nachfolger von Illinois Jacquet in Lionel Hamptons Orchester. Seine Version von Flying Home No. 2 mit Hamptons Band wurde ein Hit.

1947 gründete Cobb eine eigene Band, jedoch wurde seine Laufbahn im folgenden Jahr durch eine Operation und durch einen schweren Autounfall 1956 unterbrochen. Erst 1959 begann er wieder aufzutreten; in den 1960er Jahren leitete er verschiedene eigene Bands in Texas. Seit 1973 spielte er wieder verstärkt Aufnahmen ein und unternahm größere Tourneen und arbeitete noch 1988 in Europa mit Jimmy Heath und Joe Henderson. 1973 erhielt er eine Grammy-Nominierung als bester Jazz-Instrumentalist.

Arnett Cobb, dessen robuste Spielweise an Herschel Evans und Illinois Jacquet erinnert, ist außerdem auf Schallplatten von Herbie Fields, Hampton, Wynonie Harris und Roy Milton zu hören, außerdem auf einer Reihe von Einspielungen unter eigenem Namen (Wikipedia).
